The Skin’s Protector

Ultraviolet (UV) radiation damages skin through a variety of mechanisms, including the generation of free radicals. Gluconolactone is a polyhydroxy acid (PHA) that is capable of chelating metals and may also function by scavenging free radicals, thereby protecting skin from some of the damaging effects of UV radiation. With Advancing age, changes such as wrinkling and sagging occur to exposed skin. These changes are primarily caused by long-term sun exposure and not the mere passage of time. Clinical Study of Gluconolactone

Two types of experiments were performed. First, to determine whether gluconolactone protects against UVB-induced promoter activation in vitro, solutions containing 2.5%, 5.0%, 7.5%, and 10.0% gluconolactone in phosphate-buffered saline (PBS) was added to culture dishes for 15 minutes, followed by exposure to 5 mJ/cm2 of UVB. Gluconolactone concentrations were selected based on previous studies to determine the maximum tolerated concentrations of gluconolactone. Control cells received no UVB or gluconolactone. UVB control subjects received 5-mJ/cm2 UVB without receiving gluconolactone. Gluconolactone controls received 2.5%, 5.0%, 7.5%, and 10.0% gluconolactone and no UVB.

Observation of the Studies

Gluconolactone protects against the UV-induced increase in elastin promoter activity. Fibroblasts, derived from skin of transgenic mice containing the human elastin promoter/CAT construct, were incubated with shown concentrations of gluconolactone and then exposed to 5 mJ/cm2 of UVB. Untreated control cells received no gluconolactone. Open rectangles are untreated fibroblasts (Cont) and fibroblasts treated with 2.5%, 5.0%, 7.5%, and 10.0% of gluconolactone and receiving no UVB. Overall, gluconolactone administered 15 minutes before fibroblast irradiation afforded up to 50% protection against UVB.

Study Results

The PHA gluconolactone protects against UV radiation in vitro, as measured by the transgenic model of cutaneous photoaging. Although gluconolactone afforded some protection in the in vitro system by its ability to block the shorter wavelengths of UV, the UV absorbance spectrum shows that gluconolactone provided approximately 50% protection from elastin promoter induction. Because gluconolactone UV absorption is so poor, its protective effect must be due to mechanisms other than direct absorption of UV, such as its ability to function as a chelating agent. This ability in turn results indirectly in its ability to scavenge free radicals and protect against photoaging. PHAs offer an alternative to AHAs and retinoids in the treatment of photoaging and may offer some benefits relative to sun sensitivity. In general, PHAs are gentler to the skin than the AHAs while still serving to reduce the signs of photoaging.
